I've been replaying this game recently, and I came up with a few challenges.

1. Full Speed Up Challenge-- You can use any powerup, but you have to have five speed ups by the end of the stage you're on.

2. Pacifist Challenge-- You can only shoot bosses and enemies in the transition areas, but you CAN ram enemies with your shields (OOPS RAN OVER A CAT)

3. Moption-up challenge-- After the intro area, you can only use the missile, option, and speed up. If you die, you have to upgrade to the missile by the end of the stage.

4. No-options-- pretty straightforward. You can get every upgrade except for the option.

I bet these challenges could be ported to other Gradius games, maybe even Parodius.

Now all I need are the skills to beat any Gradius normally
